چکیده

BACKGROUND: Lovastatin is known to inhibit its own synthesis in the fungus Aspergillus terreus. Therefore, the use of a fermentation strategy that continuously removes some of the lovastatin produced from the bioreactor can enhance its productivity. This paper reports on the effects of dilution rate and the composition of the feed medium on lovastatin production by A. terreus. RESULTS: The feeding strategy consisted of an initial batch/fed-batch phase and a semi-continuous culture phase in which the pelleted biomass was retained inside a slurry bubble column. A nitrogen-free medium was fed at various fixed dilution rates in the semi-continuous phase. In experiments that were designed to assess the effects of the composition of the medium, the dilution rate was held at 0.42 d−1, but different feed media were used in separate runs. The best two-staged production strategy was shown to consist of a 96 h batch/fed-batch phase that used a nutritionally complete medium. This was followed by a semi-continuous operation using a medium that was free of both nitrogen and carbon sources. CONCLUSION: Semi-continuous operation enhanced productivity of lovastatin by 315% compared with a conventional batch operation. The optimal dilution rate in semi-continuous operation was about 0.42 d−1.  2008 Society of Chemical Industry

Lovastatin Production by Aspergillus terreus Using Agro-Biomass as Substrate in Solid State Fermentation

Ability of two strains of Aspergillus terreus (ATCC 74135 and ATCC 20542) for production of lovastatin in solid state fermentation (SSF) using rice straw (RS) and oil palm frond (OPF) was investigated. Results showed that RS is a better substrate for production of lovastatin in SSF. Maximum production of lovastatin has been obtained using A. terreus ATCC 74135 and RS as substrate without additi...

Production of lovastatin and itaconic acid by Aspergillus terreus: a comparative perspective

Aspergillus terreus is a textbook example of an industrially relevant filamentous fungus. It is used for the biotechnological production of two valuable metabolites, namely itaconic acid and lovastatin. Itaconic acid serves as a precursor in polymer industry, whereas lovastatin found its place in the pharmaceutical market as a cholesterol-lowering statin drug and a precursor for semisynthetic s...

Complete Genome Sequence of Soil Fungus Aspergillus terreus (KM017963), a Potent Lovastatin Producer

We report the complete genome of Aspergillus terreus (KM017963), a tropical soil isolate. The genome sequence is 29 Mb, with a G+C content of 51.12%. The genome sequence of A. terreus shows the presence of the complete gene cluster responsible for lovastatin (an anti-cholesterol drug) production in a single scaffold (1.16).

Screening of Lovastatin Production by Filamentous Fungi

In the present study, 110 fungal strains of Persian Type Culture Collection (PTCC) including some selected strains isolated in various screening projects were tested for their potentiality to produce lovastatin, a competitive inhibitor of 3-hydroxy-3-methylglutaryl-CoA reductase (HMG-CoA reduc-tase), the rate-limiting enzyme of cholesterol biosynthesis. The fungal strains were cultivated in a t...

Characterization of Lovastatin biosynthetic cluster proteins in Aspergillus terreus strain ATCC 20542

Aspergillus terreus is a filamentous ascomycota, which is prominent for its production of lovastatin, an antihypercholesterolemic drug. The commercial importance of lovastatin with annual sales of billions of dollars made us to focus on lovastatin biosynthetic cluster proteins. The analysis of these lovastatin biosynthetic cluster proteins with different perspectives such as physicochemical pro...
